Abstract

Immature leaflet explants of Cicer arietinum L. cv Pusa-256 developed organogenic calli on MS basal medium supplemented with 2,4-D (1-25 �M) and BAP (10 �M), such calli on transfer to 5, 10 �M of BAP and TDZ along with 0.1 �M IBA produced shoots in vitro. Thidiazuron (TDZ) (10 �M) + IBA (0.1 �M) appeared better for shoot regeneration from the callus, as compared to BAP + IBA. Addition of spermidine (1-10mM) to the shoot differentiating media containing TDZ and IBA enhanced the number of shoots by 73.8%, whereas, Putrescine and potassium phosphate could not enhance the shoot organogenesis. It appears that spermidine significantly enhance the in vitro shoot organogenesis via a callus derived from immature leaf in the presence of TDZ and BAP.
